Okay, it sounds to me like you might have the "repeat one" button accidentally pressed on your VLC media player controls. (This wouldn't really be related to the Sampler extension.)

In this picture, it's the second button from the right. It has three positions:
  • don't repeat
  • loop all tracks over and over
  • loop same track over and over (superimposes a "1" on its icon, this might be what you have)
I tested mine with this "repeat one" function enabled and it seemed to behave as you describe. Maybe check that?

No, each of the three positions get me the same result.

Obviously, your button is stuck. Please start the program and then pick up your computer and slam it vigorously on a hard surface, such as a desktop, until the button comes unstuck. If that doesn't work, please apply a screwdriver, chisel, or other pointy-ended tool into the crease along the edge of the button on your screen. That should solve it.

I'm sorry then; not sure what is wrong with your VLC. The only other thing I might suggest is resetting all your preferences back to default, but only do that if you don't mind just trying it as an experiment. (I don't know if you've knowingly customized very many settings.)

It's pretty simple to reset:
